In the settings panel and under the voice tab for vent is a testing facility. Try that: Select a codec and hit test. If you're getting bars on the right, it's not a problem with your hardware set up. If you don't get bars:
Have you tried using DirectSound (or not if you are)? Are you sure that the correct settings are selected for your Hardware input mixer? Mine are Mixer: Microphone (Headset) Mux: Master Volume Line: Master Volume. Yours may differ but shouldn't significantly.
Are your line volume and outbound amplifiers set to a reasonable level? If you click the Recording (Input) button in the bottom right does it look as though the right hardware is selected. If you're using a soundcard it ought to appear there as either microphone or line in and if you're using windows 7, ought to have a green 'tick' to indicate that it's enabled as the default device.

Good chance your soundcard is just broken then.. I had some troubles with my soundcard (see few topics below), but i got them fixed by getting a new soundcard..
Only costed me 10 euro;s and all problems are fixed now :)

I experienced that onboard soundcards are usually the first things to break, so I suggest you try out a new soundcard. And like i said it doesnt cost you much, so its definatly worth the try.
